Decidual-Like NK Cell Polarization: From Cancer Killing to Cancer Nurturing

نویسندگان

چکیده

Abstract Summary: Natural killer (NK) cells accumulate at the fetal–maternal interface and represent 70% of immune in decidua (dNK) first-trimester pregnancy; they are immune-tolerant toward semiallogenic fetus “nurturing” nonkilling NK cells. A subset patients with cancer have features common dNK, which include expressing CD56, CD9, CD49a, CXCR3, being poorly cytotoxic proangiogenic, mimicking decidual nurturing role. In oncologic patient, several factors can “decidualize” cells, turning them into immune-suppressant, growth-promoting proangiogenic Here, we suggest ways to sharpen their blunted blades intercept curb cancer-nurturing attitudes restore capabilities.

Investigation of NK Cell Population in Peripheral Blood and Tumor Lesions of Patients with Breast Cancer

Background: Alteration in peripheral blood lymphocytes (PBLs) is usually investigated to provide an evidence of the host immune responses to tumor antigens. The tumor infiltrating NK cells interact most closely with the tumor cells and more accurately reflect tumor host interactions. Objective: To analyze peripheral blood and tumor associated Natural Killer (NK) cells in patients with breast ca...

Virus fuels NK cell killing of leukemia.

NK cell transplants have emerged as a potentially powerful approach to treating acute myeloid leukemia. Approaches that could be applied to enhance the NKmediated killing of leukemic cells would therefore have direct clinical importance. Oncolytic viral therapies have also shown promise in the treatment of a variety of solid cancers, leading to the recent approval of the oncolytic HSV talimogen...

Cytokines reinstate NK cell-mediated cancer immunosurveillance.

In healthy individuals, cells that lose expression of MHC class I molecules are quickly targeted for elimination by NK lymphocytes. A paradox in cancer immunology is the observation that many tumor cells often have a drastic reduction of MHC class I molecules, yet these cells are not eliminated by NK cells, as they should be. In this issue of the JCI, Ardolino et al. demonstrate that NK cells t...

Editorial: NK Cell-Based Cancer Immunotherapy

Specialty section: This article was submitted to NK Cell Biology, a section of the journal Frontiers in Immunology NK cell-based cancer immunotherapy Innate and adaptive immunity cooperate to eliminate tumors. However, when infrequent cancer cell variants are not destroyed, tumor growth and immunosurveillance enter into a dynamic equilibrium until cancer cells evade the immune system, at which ...

Conversion of peripheral blood NK cells to a decidual NK-like phenotype by a cocktail of defined factors.

NK cells that populate the decidua are important regulators of normal placentation. In contrast to peripheral blood NK cells, decidual NK (dNK) cells lack cytotoxicity, secrete proangiogenic factors, and regulate trophoblast invasion. In this study we show that exposure to a combination of hypoxia, TGF-β1, and a demethylating agent results in NK cells that express killer cell Ig-like receptors,...
